为了高效制备玉米肽,对玉米蛋白粉进行超声预处理,以多肽得率为评价指标,从Protamex、Alcalase、Flavourzyme、Protex.7 L、胰蛋白酶、木瓜蛋白酶等6种蛋白酶中筛选出Alcalase为最佳水解用酶。通过单因素试验和响应面分析确定制备玉米肽的工艺条件为:底物浓度10.0 g/(100 ml)、加酶量2 830 U/g、反应pH9.0、反应温度58.0℃、反应时间3 h。在该条件下制备的玉米酶解产物的多肽得率为(46.61±0.37)%,与理论预测值的相对误差在±1%以内。

In order to prepare corn peptides efficiently, corn gluten meal was pretreated by ultrasonic. Using the yield of peptides as the evaluation indicator, Alcalase was screened out from the following six kinds of protease as:Protamex, Alcalase, Flavourzyme, Protex.7L, Trypsin and Papain. Single factor experiment and response surface methodology were used for optimizing the enzymatic hydrolysis conditions. The optimum hydrolysis conditions were designated as substrate concentration 10.0 g/(100 ml), enzyme concentration of 2 830 U/g, pH9.0, enzymolysis temperature of 58.0℃ and enzymolysis time of 3 h. Under these conditions, the yield of corn peptides could reached up to (46.61±0.37)%,and the theoretical prediction relative error values within ±1%.
